چکیده

Regulatory T (Tr) cells have been shown to arise in the periphery during induction of peripheral tolerance. However, the mechanism involved remains elusive. In the present study, we investigated the potential role of transforming growth factor (TGF)-b in the peripheral induction of regulatory phenotypes in the conventional CD4CD25 T cells. Upon priming in the presence of TGF-b, there was greatly enhanced expression of CD25, cytotoxic T lymphocyte-associated antigen 4 (CTLA-4), and the natural Tr cell-specific transcription factor Foxp3 in naive CD4CD25 T cells. The CD25 cells that emerged later only in the TGF-b-treated culture failed to express CD69, so distinguishing this population from activated CD25 effector cells. The TGF-b-treated T cells entered an anergic state following restimulation, as judged by enhanced induction of programmed death (PD)-1, as well as impaired responses in terms of proliferation and IL-2 production. Importantly, the TGF-b-costimulated CD4CD25 T cells, prior to conversion to CD25 cells, were able to suppress the proliferation of responder T cells via contact-dependent and interleukin-10-independent mechanisms. Taken together, these results demonstrate that the existence of TGF-b during early phase of priming is sufficient to induce CD4CD25 Tr cells with anergic and immunoregulatory activities equivalent to thymus-derived CD4CD25 Tr cells, and these cells are programmed to be CD25 cells under the prolonged resting conditions. Thus, our findings provide a novel mechanism by which TGF-b mediates infectious tolerance in the periphery.

Effect of Transforming Growth Factor-β (TGF-β) on proliferation of gastric epithelial cells in culture

Objective: Helicobacter pylori has a well-established role in the development of gastric cancer. In vitro studies reveal increased proliferation of the gastric mucosa in the presence of H. pylori infection. It has been also shown that production of some cytokines, such as interleukin-1 beta (IL-1b) is in...

Suppressor activity and potency among regulatory T cells is discriminated by functionally active CD44.

CD4CD25(+) regulatory T cells are fundamental to the maintenance of peripheral tolerance and have great therapeutic potential. However, efforts in this regard have been hampered by limiting cell numbers in vivo, an anergic phenotype in vitro, and a rudimentary understanding of the molecular basis for the functional state of CD4CD25(+) regulatory T cells (Treg cells). Here we show heterogeneity ...

Regulatory CD 4 + CD 25 + cells reverse imbalances in the T cell pool of bone marrow transplanted TG e 26 mice leading to the prevention of colitis

Background and aims: Erroneous thymic selection of developing T lymphocytes may be responsible for the expansion of self reactive T cells or may contribute to the absence of regulatory T cells important in controlling peripheral inflammatory processes. Colitis in bone marrow (BM) transplanted Tge26 mice is induced by abnormally activated T cells developing in an aberrant thymic microenvironment...

Human CD25+CD4+ T Suppressor Cell Clones Produce Transforming Growth Factor β, but not Interleukin 10, and Are Distinct from Type 1 T Regulatory Cells

T regulatory (Tr) cells are essential for the induction of peripheral tolerance. Several types of Tr cells exist, including CD4(+) T cells which express CD25 constitutively and suppress immune responses via direct cell-to-cell interactions, and type 1 T regulatory (Tr1) cells, which function via secretion of interleukin (IL)-10 and transforming growth factor (TGF)-beta. The relationship between...
